Cymru Alliance winners Prestatyn will be promoted to the Welsh Premier next season at the expense of Llangefni.
Second-placed Bala had also applied for promotion, but when Prestatyn passed the league's ground inspection their elevation was secured.
"Prestatyn are a progressive club," said league secretary John Deakin.
No team will be promoted from the Welsh League as their top two sides did not apply for elevation, meaning that Caersws gain a Welsh Premier reprieve.
Prestatyn have never appeared in the Welsh Premier before and, at the age of 28, their manager Neil Gibson will be the youngest in the league.
Neath Athletic have secured their Welsh Premier future by leaving Llandarcy Park to ground-share the Gnoll with Neath RFC.
